There are 2 puzzles in the Director's Office - the hand puzzle and the hieroglyphics puzzle. Once you get to Brookhaven Hospital, you'll find the Director's Storage Room Key early on, but it will be on the last places you unlock in that area.
By following the main story, you'll eventually end up in the Director's Storage Room, with a hand sitting on the Director's desk. You'll be tasked with finding 3 bracelets for the Hand which will make it open and give you the key to the Director's Office.
The Marked Bracelet can be found after entering room L1. James will fall through a hole between the walls, and you'll end up in the Medical Records area, and the Marked Bracelet will be in the back corner.
The Filthy Bracelet is in Room D1 on floor 3F. To gain access to Room D1, you'll have to solve the x-ray viewer puzzle. Here's how the radiographs should be arranged for the X-ray viewer puzzle:
The Bloodstained Bracelet is located in the Pool on Floor 1F. To access it, you'll need to combine several tools you've found along the way to drain the water. Combine the Medical Tube and Bent Needle to access the Pharmacy. From there, you can get the Pool Pump. Drain the pool to find the Bloodstained Bracelet.

Stack the bracelets like this from top to bottom, and the hand will open to give you the key to the Director's Office, where we finally encounter the last safe.
The safe in the Director's Office requires arranging some books with hieroglyphics on them, and they need to be arranged in a specific order. Arrange the books to complete the lion like this:
After arranging the books, return to the safe and use the numbers on the Bracelets to unlock the safe. The code is 92, 45, 71. The solution will look like this:
